【Academic Report】The 106th "Chen Bangjie Forum": Genetic Engineering Empowers the Next Generation of Cellular Immunotherapy
On the afternoon of December 26, 2025, the 106th session of the "Chen Bangjie Forum" was successfully held in the sixth-floor conference room of Nanjing Miracle Biotech. At the invitation of Professor Guo Zhigang from the School of Life Sciences, Nanjing Normal University, Professor Ye Lupeng, a National Outstanding Young Scholar and doctoral supervisor at the Institute of Modern Biology, Nanjing University, visited for academic exchange and delivered a wonderful academic report entitled Immune Cell Engineering and Cellular Immunotherapy. The report was chaired by Professor Guo Zhigang and was enthusiastically attended by faculty and students of the school.

Based on this revolutionary tool, Professor Ye further shared how his team used the AAV-SB-CRISPR in vivo screening technology to systematically identify key genes regulating immune cell function in the authentic tumor microenvironment. He detailed two breakthrough discoveries: first, the identification of membrane protein genes such as PDIA3 and MGAT5, which act as intracellular "brakes" in T cells. Knocking out these genes in CAR-T cells can significantly enhance their efficacy against solid tumors. Second, the metabolic enzyme gene PRODH2 was discovered as an "energy amplifier" for T cells, which can greatly improve the persistence and anti-tumor activity of CAR-T cells by reprogramming cellular metabolism. These findings provide novel targets and strategies to overcome the core bottlenecks currently facing cellular immunotherapy, including insufficient persistence and poor infiltration into solid tumors.

This report offered faculty and students a valuable opportunity to communicate face-to-face with a leading young scholar in the fields of gene editing and cell therapy. The research paradigm of "tool development-driven biological discovery" broadened the academic horizons of all attendees and greatly inspired in-depth thinking on immune engineering and translational medicine.
